2025 Annual Report
Meeting the Moment
ALS research has reached a turning point. New insights into genetics and disease biology, alongside advances in biomarkers and artificial intelligence, are giving researchers more credible therapeutic ideas to pursue than at any point before. However, identifying those ideas is only the beginning. Every potential idea has to be rigorously tested and advanced through the stages of drug discovery. This is the hard but critical work that ALS TDI carries out in our lab every day.
In 2025, we continued that work while building on our capabilities. Several therapeutic programs moved forward, the ALS Research Collaborative (ARC) Study grew, and we opened new collaborations while sharing data and research tools with scientists around the world.
What follows is what that work produced in 2025, and evidence that the capabilities we've built over more than two decades are starting to open doors that weren't there before.

For many years, one of the greatest challenges in ALS research was a lack of promising therapeutic opportunities.
Today, ALS research has reached a turning point. Advances in genetics, biomarkers, artificial intelligence, and disease biology are generating more promising therapeutic opportunities than ever before. For the first time, scientific opportunity is advancing faster than the systems designed to evaluate it.
That shift demands a new approach.
Discovering a promising therapy is only the beginning. Moving it toward patients requires rigorous testing, reliable biomarkers, relevant disease models, and the ability to make evidence-based decisions at every stage of development.
This is the challenge ALS TDI was built to address.
Throughout 2025, we strengthened the capabilities that make this work possible. We advanced therapeutic programs, expanded the ALS Research Collaborative (ARC), launched Champion Insights to better understand why certain populations face a higher risk of developing ALS, and built new collaborations across academia, industry, and nonprofit organizations. Each of these efforts contributes to the same goal: giving researchers better tools, better data, and stronger evidence to move promising therapies forward.
None of this would be possible without our community. Every participant, donor, volunteer, advocate, and partner has played a role in the progress reflected in these pages.
The work highlighted throughout this report also laid the foundation for an important milestone. In early 2026, ALS TDI announced a landmark expansion of our laboratories and research capabilities. This investment reflects our commitment to meeting this moment— ensuring we have the capacity to evaluate more therapeutic opportunities, advance the strongest ideas with rigor, and help bring effective treatments to people living with ALS faster.

Every program begins with the same question

Every program begins with the same question: could this approach lead to a meaningful treatment for people living with ALS?
There is no single cause of ALS, and there won't be a single treatment that works for everyone. That's why ALS TDI is pursuing multiple therapeutic strategies while developing the research tools needed to evaluate them.
Throughout 2025, our researchers advanced new therapeutic approaches while continuing to develop better disease models and biomarkers. Together, these efforts strengthen every stage of the drug discovery process, from identifying promising therapies to evaluating them in preclinical testing and clinical trials.

Advancing ALS Treatments icon
Advancing Treatments for ALS

Messenger RNA (mRNA) has changed how scientists approach the treatment of many diseases. In 2025, ALS TDI began pioneering this technology for ALS by partnering with Dr. Michael Mitchell at the University of Pennsylvania to develop lipid nanoparticles designed to deliver mRNA-based therapies to the central nervous system. Our scientists also synthesized several mRNA candidates for preclinical testing. Over the next year, we'll be evaluating whether these mRNA candidates can effectively reach and act on key therapeutic targets in ALS.

In 2020, ALS TDI researchers discovered that type I PRMT inhibitors can alter ALS-linked proteins associated with the C9orf72 mutation, the most common genetic cause of ALS. Building on that discovery, our team is now testing PRMT inhibitor compounds in animal models to  determine whether they can correct RNA mis-splicing and reduce protein aggregation—two biological processes believed to contribute to disease progression. Although this work began with C9orf72-related ALS, researchers believe the biology may ultimately have broader applications across other forms of the disease.

Research into copper biology also continued to move forward in 2025. TDI-1831, ALS TDI's most advanced therapeutic candidate, advanced into IND-enabling studies. At the same time, researchers developed patient stratification strategies to help identify the people most likely to benefit from this approach. These studies are generating the data needed to  determine  whether TDI-1831 is ready to move into clinical testing.

Building Better Ways to Test Therapies
Discovering a promising therapy is only part of the challenge. Researchers also need reliable ways to evaluate whether that therapy is having the intended effect.
Profilin1 Mouse Model: ALS TDI researchers published the first comprehensive characterization of a Profilin1 mouse model, demonstrating that it closely reflects key features of ALS. This work provides researchers with another well-characterized model for evaluating potential therapies before they move into clinical testing.

Measuring What Matters
Biomarkers help researchers measure how ALS progresses and whether an experimental treatment is having its intended effect. Better biomarkers lead to better clinical trials.
Digital Biomarkers:  Researchers continued evaluating how voice recordings and wearable device data collected through the ARC Study correlate with clinical outcomes, bringing these digital biomarkers closer to use in ALS clinical trials. 
Omics:  Analysis of omics data from ARC participants expanded in 2025, helping identify biological signatures associated with disease progression and treatment response.
C9 Biomarkers:  The team also developed new antibodies against methylated C9orf72 dipeptide repeats, research tools that may support future biomarker development and therapeutic research. 
As these tools continue to improve, they have the potential to make clinical trials more informative, more efficient, and better able to identify therapies that truly benefit people living with ALS.
The Power of Research Participation

ALS takes many different forms, and researchers can only make sense of that variation with direct participation from the ALS community. When participants share health information, biological samples, and their experiences with ALS, they give researchers a clearer view of how the disease differs from one person to another and how it changes over time.
In 2025, ALS TDI's partnership with the ALS community continued through the growth of the ALS Research Collaborative (ARC) Study and the announcement of Champion Insights.

The ALS Research Collaborative (ARC)
The ALS Research Collaborative (ARC) is the longest-running natural history study in ALS and one of the largest efforts to collect real-world data from people living with the disease.
Through ARC, participants contribute health records, survey responses, voice recordings, wearable device data, and blood samples.
Each data type helps researchers better understand how ALS develops and progresses. Those data are shared through the ARC Data Commons, where researchers around the world can use them to ask new questions, test new ideas, and build on one another's work.

One of the world's largest repositories for ALS digital biomarker research
Behind those numbers are thousands of people who have contributed their data to move ALS research forward. Here are a few ways researchers are putting ARC data to work today.

Together with Axol Bioscience, ALS TDI is developing stem cell models that better reflect the diversity of ALS, with an emphasis on development of sporadic ALS models. Many of these models are created using samples donated by ARC participants, giving researchers more representative tools for studying the disease and evaluating potential therapies.

ARC data is also supporting the Longitude Prize on ALS, a global initiative that uses artificial intelligence to uncover patterns hidden within large research datasets. By combining ARC data with other sources, researchers hope to identify new clues about how ALS develops and where new treatment opportunities may exist.

ARC data is being added to the Answer ALS Neuromine platform, where researchers will be able to access and explore it alongside other major ALS datasets. Bringing these data together will make it easier for scientists to compare findings, investigate new research questions, and work across institutions.

Champion Insights: Understanding Risk to Find New Answers
Military veterans, elite athletes, and first responders seem to be diagnosed with ALS at higher rates than the general population. Champion Insights was created to investigate why.
Announced in 2025 by ALS TDI, Answer ALS, and Augie's Quest to Cure ALS, the study will investigate the genetic, biological, clinical, environmental, and lifestyle factors that may contribute to the increased risk of ALS in these populations.
Champion Insights builds on the foundation of the ARC Study. By comparing data from these higher-risk groups with the broader ARC population, researchers will be able to identify patterns, evaluate potential biomarkers, and better understand the biological pathways that may contribute to ALS.
The insights gained from this work could lead to new approaches for understanding, diagnosing, and ultimately treating the disease.

ARC now includes contributions from thousands of participants across 50 countries. Champion Insights adds a new, targeted layer to that dataset — and each new participant sharpens what both studies can reveal.
As these data sets continue to grow, so does our ability to advance more effective treatments for ALS.
Driving Progress Through Collaboration

No single organization has the resources to identify, test, and validate every promising approach to treating ALS. Progress requires researchers, nonprofits, and industry partners to share data and expertise — and it requires an organization positioned to carry promising discoveries from the lab toward the clinic.
That is the role ALS TDI was built to play.
ALS TDI has built the scientific team, research platforms, and infrastructure to do that work. Our team evaluates therapeutic approaches from outside labs and industry partners against our own data and models to determine which have the greatest potential to advance. In 2025, several collaborations strengthened this work.

2025 Collaboration Highlights

ALS TDI partnered with Axol Bioscience to develop and distribute stem cell models derived from people living with sporadic ALS. Because they're built from samples donated by ARC participants, these models capture genetic and clinical diversity that most existing ALS models lack. This work was an integral part of the PRISM initiative, which was announced in 2026 to expand access to high-quality stem cell models that better reflect the biological complexity of ALS.

Together with Answer ALS and Augie’s Quest, ALS TDI announced the launch of Champion Insights, a first-of-its-kind research initiative focused on understanding why military veterans, first responders, and elite athletes appear to face a higher risk of developing ALS. Built on the ARC platform, the study will generate new data that could reveal important clues about disease risk and biology.

Working with LifeArc, ALS TDI is identifying and validating blood-based biomarkers that could improve how ALS is diagnosed and monitored. Using samples collected through the ARC Study, researchers have already identified promising markers that may help predict how quickly the disease progresses—an important step toward improving clinical trials and evaluating future therapies.

Through a collaboration with the Critical Path Institute (C-Path), ALS TDI is evaluating wearable device data from the ARC Study to identify potential new ways to measure disease progression in ALS clinical trials. These digital biomarkers could help make future trials more sensitive and better able to detect whether a potential treatment is having a meaningful impact.

The Longitude Prize on ALS, organized by Challenge Works, challenges researchers around the world to develop AI-powered tools that accelerate ALS research. As a data partner, ALS TDI is contributing more than a decade of patient data from the ARC Study. We’re also collaborating with researchers at Google to explore how artificial intelligence can uncover patterns that may lead to new insights into ALS.
Advancing Research Together
These collaborations reflect the different roles ALS TDI plays within the ALS research ecosystem. Whether we're providing data, developing models, validating discoveries, or evaluating new therapies, the goal is to move the most promising ideas forward.
ALS won't be solved by one lab or one breakthrough. But by bringing together the right people, tools, and evidence, we can move faster toward treatments that work.

At ALS TDI, we believe that sharing findings with the global research community is essential to advance our collective understanding of ALS. In 2025, ALS TDI researchers published papers, presented posters, and spoke at conferences across the country, including our own ALS TDI Summit in Boston.

At the 2025 ALS TDI Summit, our researchers and collaborators presented the year's work — from new technologies to shared datasets to the partnerships described above — and discussed how this shapes where ALS drug discovery goes next. The day's agenda included:

Community Impact
As a nonprofit biotech, ALS TDI depends on our community to fund and drive every research breakthrough. In 2025, more than 200 individuals organized fundraisers for ALS drug discovery, joined by thousands of supporters who donated, volunteered, and spread the word.

Brewing a Cure Goes Golfing
~$300,000 raised in 2025
In May of 2018, Dennis Ceruti—a hardworking, healthy living, loving husband and father of four amazing girls—was diagnosed with ALS. The next year, his friends and family organized Brewing a Cure for ALS, a beer festival that raised nearly $150,000 for ALS TDI.
Sadly, Dennis passed in May of 2020 after a 2-year battle with ALS. It was always his goal to raise money to find a cure, to ensure no one else, or their families had to endure the pain that is ALS. In 2024, led by two of Dennis’s best friends, Camille and Dave, friends and family of Dennis came together and re-lauched Brewing a Cure for ALS as a golf outing. In 2025, year two of Brewing a Cure Goes Golfing, nearly $300,000 has been raised in honor of Dennis for research at ALS TDI.

The Hummingbird Fund

The Hummingbird Fund was founded by Drs. Jim and Peggy Plews-Ogan along with their family and friends. It serves as an accelerator for innovation in ALS research, advocacy, and overall functional support, through stable and sustainable partnerships that are family- and patient-centric. Sadly, Jim passed away on July 3, 2024, but the work of the Hummingbird Fund continues, in Jim’s honor and memory.
Following their mission to support patient-centric research, The Hummingbird Fund has generously donated over $100,000 (over 2 years), to support the work at ALS TDI, specifically our ARC study. Their support has helped us to increase enrollment in the ALS Research Collaborative (ARC) Study, with a focus on enrolling young women who have been diagnosed with ALS.

Dance Away ALS

Dance Away ALS is an annual evening of music, dancing, and raffles held at Noble Funk Brewing Company in Louisville, Kentucky. It is organized by Katie Rixman, who lost her sister, Susanna Dennis, to the disease.
In 2025, the event honored Katie's friends living with ALS today, Stephan Korner and Aimee Smith, as well as those she has lost to the disease, including her Susanna, Bill Cunningham, Terry Hunt, Laura McDonald, Lyle Hawkins, Hal Burgiss, Tonya Anderson, Randy Puno, and many more. Over three years, the event has raised more than $145,000 for ALS research at ALS TDI, including $56,000 in 2025.

Pedals vs. ALS
$50,000+ raised
On May 1, 2025, Timm Fenton started riding his bike east from the Pacific Coast in Florence, Oregon. 67 days later he arrived at the Atlantic Ocean in Yorktown, Virginia.
Timm's effort, which he named "Pedals vs. ALS," was inspired by his mother, Jan, who he lost to ALS in 2017. Along his more than 4200-mile route, he crossed snow- covered mountains, faced intense storms through the Great Plains, and was even chased by wild dogs on remote country backroads. Timm shared the stories of his triumphs and challenges, as well as spreading ALS awareness, throughout his journey, ultimately raising more the $50,000 for ALS research at ALS TDI.

Across ALS research, there are now more promising leads than the field has the capacity to test.
ALS TDI announced a landmark expansion of its lab and research team, beginning in 2026. This investment will allow us to evaluate more therapies, develop more advanced disease models, strengthen biomarker discovery, and move the most promising programs forward more efficiently.

Augie's Quest to Cure ALS: A Commitment Built Over Time
Augie's Quest to Cure ALS has supported ALS TDI for nearly two decades. Over the years, that partnership helped ALS TDI strengthen its research capabilities and build the foundation for this next stage of growth.
Today, a transformational commitment from Augie's Quest to Cure ALS is helping make ALS TDI's landmark expansion possible.
This commitment carries forward the vision of Augie Nieto, co-founder of Augie's Quest to Cure ALS and longtime chair of ALS TDI's Board of Directors. Augie understood that ending ALS would require research conducted with urgency, ambition, and the flexibility to follow the science wherever it leads.

Expanding Our Research Capacity
Following the announcement of our expansion, we will begin growing our science team and begin construction on a new research facility in Watertown, MA. By doubling the size of our lab and our team, we'll be able to explore more ideas and test more promising drugs in parallel.
Expanding Research Participation and Data Resources
The ALS Research Collaborative (ARC) Study will continue to grow as one of the world's most comprehensive ALS natural history studies. At the same time, Champion Insights will begin enrolling military veterans, elite athletes, and first responders, with the goal of uncovering factors that may contribute to higher risk of ALS in these populations.
Advancing Therapeutic Discovery
Our researchers will continue advancing multiple therapeutic programs, including mRNA technologies, TDI-1831, and PRMT inhibitors. At the same time, we will continue to pursue new therapeutic opportunities emerging from discoveries in our own lab and from collaborations across the ALS community.
Building Better Tools for Drug Discovery
ALS TDI will continue expanding the tools researchers need to develop better therapies. That includes launching PRISM ALS to increase access to sporadic models of ALS, advancing digital and blood-based biomarkers, and improving the technologies that help researchers evaluate potential treatments.
